چکیده
Title of Document: A NEW INSIGHT INTO MYCOBACTERIUM RESISTANCE TO REACTIVE OXYGEN INTERMEDIATES. Rabab Kish, Master of Science, 2006 Directed By: Dr. Lian-Yong Gao, Department of Cell Biology & Molecular Genetics The genes encoded by the RD1 locus are known to be important for intracellular survival of pathogenic mycobacterium, however their role in counteracting host defense is not known. I hypothesize that RD1 is involved in counteracting host oxidative response by secreting ROI-neutralizing enzymes such as catalase and superoxide dismutase. In support of this hypothesis I have shown that M. marinum RD1 mutants are more sensitive than wild type to ROIs both in vitro and in BMDM. Western blot analysis on the KatG protein levels within the bacterial cells grown in 7H9 rich medium demonstrated that wild type bacteria produced significantly higher amount of KatG than the RD1 mutants. When the bacteria were exposed for 2 hours to H2O2, wild type showed a significant reduction of the KatG level, while the RD1 mutants maintained constant levels of KatG, suggesting that RD1 genes might be involved in the secretion of KatG upon exposure to H2O2. These results together demonstrate an important unknown function of RD1 in resistance to ROIs. A NEW INSIGHT INTO MYCOBACTERIUM RESISTANCE TO REACTIVE OXYGEN INTERMEDIATES

Reactive oxygen and nitrogen intermediates in the relationship between mammalian hosts and microbial pathogens.
This review summarizes recent evidence from knock-out mice on the role of reactive oxygen intermediates and reactive nitrogen intermediates (RNI) in mammalian immunity. Reflections on redundancy in immunity help explain an apparent paradox: the phagocyte oxidase and inducible nitric oxide synthase are each nonredundant, and yet also mutually redundant, in host defense. In combination, the contr...
متن کاملMycobacterium tuberculosis CDC1551 is resistant to reactive nitrogen and oxygen intermediates in vitro.
Resistance to reactive oxygen intermediates and reactive nitrogen intermediates in vitro of a clinical isolate of Mycobacterium tuberculosis (CDC1551) that caused a large outbreak of tuberculosis was compared to that of M. tuberculosis strains CB3.3, H37Rv, H37Ra, Erdman, RJ2E, C.C. 13, and C.C. 22 as well as M. bovis strains Ravenel and BCG. CDC1551 and CB3.3 were significantly more resistant ...
متن کاملReactive nitrogen intermediates have a bacteriostatic effect on Mycobacterium tuberculosis in vitro.
Susceptibility of six isolates of Mycobacterium tuberculosis (CB3.3, CDC1551, RJ2E, C.C.13, H37Rv, and H37Ra) and two isolates of Mycobacterium bovis (Ravenel and BCG) to reactive oxygen intermediates (ROI) and reactive nitrogen intermediates (RNI) was determined by standard in vitro survival assays. After 21 days of incubation, the survival of most strains exposed to either acidified sodium ni...
متن کاملCu,Zn superoxide dismutase of Mycobacterium tuberculosis contributes to survival in activated macrophages that are generating an oxidative burst.
Macrophages produce reactive oxygen species and reactive nitrogen species that have potent antimicrobial activity. Resistance to killing by macrophages is critical to the virulence of Mycobacterium tuberculosis. M. tuberculosis has two genes encoding superoxide dismutase proteins, sodA and sodC. SodC is a Cu,Zn superoxide dismutase responsible for only a minor portion of the superoxide dismutas...
متن کاملA Novel Antioxidant Gene from Mycobacterium tuberculosis
Among the major antimicrobial products of macrophages are reactive intermediates of the oxidation of nitrogen (RNI) and the reduction of oxygen (ROI). Selection of recombinants in acidified nitrite led to the cloning of a novel gene, noxR1, from a pathogenic clinical isolate of Mycobacterium tuberculosis. Expression of noxR1 conferred upon Escherichia coli and Mycobacterium smegmatis enhanced a...
